  • PhD research-title: Investigating the role of overexpressed cytochrome P450 CYP325A in pyrethroid-based metabolic resistance in Anopheles funestus, a major malaria vector across Africa.
  • Research objectives involved using molecular biology techniques (PCR, cloning and sequencing) to amplify and characterize a mosquito P450 gene (CYP325A) supervised by Dr Michael O. Kusimo.
  • In silico predictions (modelling and molecular docking simulations) using Modeller and Molegro Virtual Docker software.
  • In vitro ( heterologous expression in pCW vector of P450 in E. coli coexpressed with CPR, metabolism assays with insecticides and enzyme turnover kinetic studies) This par was conducted in the Vector Biology Laboratory of Pr. Charles Wondji supervised by Dr Sulaiman S. Ibrahim, at the Liverpool School of Tropical Medicine, United Kingdom.
  • In vivo techniques of RNA interference and transgenic expression of mosquito gene in Drosphila melanogaster flies (pUAS-Gal4 system)
  • Research duties and work were supported by a Wellcome Trust Senior Research Fellowship in Biomedical Sciences to CSW (101893/Z/13/Z and 217188/Z/19/Z) as a scholarship student.

Masters research title: Investigating the larvicidal potential of Tithonia diversifolia , a sunflower plant against Anopheles and Culicine mosquitoes in Yaounde, Cameroon.

Research duties involved:

  • Mandatory course work at the university and a minimum of 2.4GPA for the masters one level.
  • Collection of sunflower leaves and flowers and pre-treatment before extraction procedures.
  • Hydrodistillation of fresh samples to acquire essential oils for HPLC analyses of composition.
  • Cold aqueous and ethanol maceration of dried samples and performed phytochemical screening of secondary metabolites on macerates
  • Field collection of mosquitoes from larval breeding sites and rearing in the laboratory to stage 4 for bioassay.
  • Conducted larvicidal bioassay on Anopeles and culicine larvae and registered susceptibility levels through mortality rates.

  • Worked as English/French translator at meeting and for documentation to facilitate communication between stakeholders and Cameroon government officials mostly french-speaking.

  • Heterologous expression of CYP325A in E. coli involved sub-cloning of gene into pCW expression vector and co-expressing with CPR-Cytochrome P450 Reductase.
  • Harvesting protein and testing for functional protein by complexation with carbonmonoxide and reading on spectrophotometre for read at P450 if active protein. Then purification and quantification.
  • Metabolism assays conducted using pyrethroid (permethrin, deltamethrin and alpha cypermethrin) and non-pyrethroid (propoxur and DDT) insecticides for selected alleles from cameroon and DRC.
  • Enzyme turnover kinetic studies for permethrin and deltamethrin to measure catalytic efficiency of alleles.
  • This entire process involved a keen respect of laboratory protocols and proper use of complex equipment such as ultra centrifuges, HPLC machines, spectrophometer, etc.
